TESTING METHODOLOGY: High Performance Liquid Chromatography Triple Quadrupole Mass Spectrometer
(LC/MSMS).
 
SPECIMEN REQUIREMENTS:
  • Collect: 5 ml Gold (serum) or Red top tube. Separate serum (min volume required is 0.5 ml)
  • Min. Collection: 1.0 ml whole blood
  • Transport: Refrigerated
  • Stability: After serum separation. Ambient: 24 hours; Refrigerated: 1 week. Frozen: 1 month
  • Unacceptable Conditions: Room Temperature specimens older than 24 hours.
* There are no established reference ranges for 25-Hydroxy Vitamin D2 and D3 alone.
 
REFERENCE INTERVAL:
Normal ranges are based upon literature values and expert opinion (Table 1) until population specific values are attained.

Vitamin D helps the body absorb calcium, which is needed for bone growth and health. A lack of vitamin D can lead to bone diseases such as osteoporosis or rickets. Vitamin D also has a role in the nerve, muscle, and immune systems.
There are three sources of Vitamin D : through the skin after sunlight exposure, diet, and from supplements.
 
Analytic Sensitivity: >99%
Test Limitations: This methodology is not valid for children less than 1 year old. Significant concentrations of C-3 epimers of 25-OH vitamin D2 and D3 are commonly found in infants. This can lead to overestimation of total 25-OHD levels. Measurement of total 25-OHD in children less than 1 year of age will therefore not be performed by LC/MSMS at this facility. These specimens will be outsourced for analysis.
 
INDICATIONS FOR USE:
The intended purpose of this procedure is to measure total
Vitamin D levels in serum using Liquid chromatography-tandem mass spectrometry (LC/MSMS).
